Install to thumb drive with permanent drive letter

Apologies in advance if this was covered, I tried searching and came up with no answers.

Just looking to take a different tack for transportable files. Assuming a large enough portable drive for the moment, if the drive were set to a static drive letter in windows (see here for details) for each machine that I want to use RW on (e.g. home and laptop), is there anything that RW puts in the registry that would cause headaches when moving from machine to machine?
 
*DO NOT DO THIS!!*

Right now, the database is in a fixed location. Once we allow it to no longer be fixed, we will be taking steps to ensure this cannot be done. The database can NOT be shared using this technique. If you do this, bad things will happen. :(
