I am wondering is it is possible to create a custom house rule that will add the characters Constition score (as well as there Modifier) to the characters hit points at first level?

Right now I am just creating a Permanent Adjustment per character for this, but would love to be able to have a checkbox for it in the 'Configure Your Hero' page.

You can create your own source and make the parent the house rules source. If that is selected, have an eval script on something added to all heroes (I used to use Conditions) perform the addition of Con score to HP (similar to the Toughness feat).
